At the Figure 2 shows scheme of the purine-dependent Ca 2+ transduction at the
cells. The metabotropic purinoreceptors P2Y throughout the G-proteins activate PLC
that produces InsP 3 , then InsP 3 bounds with InsP 3 -receptors, that release Ca 2+ from
ER. And retrograde signal CIF (related to ER Ca 2+ store depletion) go to the cellular
surface, and the large À ow of Ca 2+ introduced to the cell throughout the Ca 2+ release-
activated Ca 2+ channels (CRAC) [24]
So, after ATP addition occur two large increasing of intracellular Ca 2+ concentra-
tions. As a result ɋɚ 2+ -dependent Ʉ + ɢ Cl - channels are activated. The volume of cells
changed, because these channels regulate the cell volume. The compensators H 2 O À ows
occurred to the cellular interiors. And cellular volumes are increased greatly and bitterly.
Than Ca 2+ concentrations are decreased quickly, Ca 2+ is attenuated by mitochondrial,
or ER or it is removed from the cell. After than Ca 2+ concentration in the cell increases
smoothly again. ɋɚ 2+ dependent Ʉ + ɢ Cl - channels are activated again. The cellular vol-
ume increase slowly, too. Thus, the light scattering of dilute suspension of EAC cells
were changed too twice. There are two large increasing of intracellular Ca 2+ concentra-
tions, as the result we obtain two maximums at the light scattering kinetic curves.
We recorded of right angle light scattering of dilute suspension of EAC cells with
PerkinElmer-44B spectrophotometer at a wavelength of 510 nm. The control samples
showed the bimodal cellular responses (Figure 3). Urgently after the ¿ rst addition of
ATP to the cell suspension the big maximum appeared quickly on the kinetic curve
(¿ rst peak). Then it bust momentary. After that the low plate is appeared. And than the
slow rise up is developing to plate, that is equal or above to the reference levels (sec-
ond peak). When we repeated the addition of ATP, the picture was repeated. Melafen
inhibited the response development by the dozes dependent manner (Figure 3).
