adhesion and expansion, MSC-loaded scaffolds can be incubated for
anywhere from an hour to several days under conditions favorable
forMSCgrowth.Intwo-dimensional(2D)cultureconditions,growth
factors, alone or in combination with dexamethasone, dramatically
enhance proliferation of MSCs. In a three-dimensional (3D) porous
ceramic scaffold, the combination of FGF-2 and dexamethasone
also significantly stimulates both MSC proliferation and osteoblast
lineage commitment. Seeding of bone marrow MSCs in a porous
biphasic calcium phosphate (BCP) scaffold, which is composed of
hydroxyapatite and β -tricalcium phosphate (6:4), followed by incu-
bation with FGF-2 and dexamethasone for 7 or 14 days, results in
enhanced growth of MSCs compared to control (Fig. 41.1). Under
