Figure 6.9
DHM investigation of Stx1-induced cell death of a single HBMEC and an EA.hy cell. (A) False
color-coded digital holographic phase contrast images of cells at indicated time after Stx1
addition. The magnification bars correspond to 10

quantitative DHM phase images whereby the x-axis matches the lines in panel (A). The parameters

( n medium 5 1.337) that was determined on suspended cells as described in Section 6.4 .
Figure 6.9C shows the temporal dependence of the maximum OPL Δϕ max and the
corresponding maximum cell thickness d max .
For the HBMEC ( Figure 6.9 , left panels), the first observed reaction was cell rounding after
20 h followed by an increase in the cell thickness by up to 50% after 35 h ( Figure 6.9C , left
panel), with simultaneously decreased intracellular fluctuations. After 45 h, the phase
contrast started to decrease as a sign of progressive cell disintegration. Finally, the cell
death, accompanied by an extensive cell leakage, occurred after , 51 h of Stx1 treatment
( Figure 6.9A , left panel). In contrast, initial Stx1-mediated cell rounding of the EA.hy 926
