The low doses of 10 -12 -10 -5 M Melafen inÀ uence to the fate of animal tumor cells
in vitro . The content of protein “labels of apoptosis”--protein regulator p53 (increase)
and antiapoptosis protein Bcl-2 (decrease), that were showed by immunoblotting
methods at the EAC cells. This fact indicated that the apoptosis was developed under
the 1, 5 hr of Melafen action. And similar dozes of Melafen in vivo suppressed the
growth of Luis carcinoma [17].
Thus, Melafen looses the structure of the soluble protein, changes the microdo-
maine organization of DMPC membrane and does not change the structural organiza-
tions of ghost and egg lecithin membranes. But Melafen global change the erythrocyte
morphology and delayed the rate of growth of solid Luis carcinoma.
Taking into account, data obtained the A. E. Arbuzov Institution of Organic and
Physical Chemistry, about formation by the Melafen in aqueous solutions of supra-
molecular structures involving of water molecules [18]. It can be assumed that just
such the structures change the microdomaines organization in attachable delicately
organized and labile structures. This assumes may be real only for interactions be-
tween biological objects with Melafen in aqua solutions. But in the cellular interior
there is not the suf¿ cient amount of free waters molecules for such supramolecular
structures formation. It is hard to suppose, that the linkage with Melafen will ¿ nd
stronger, than with nature cellular chelating agents of water. To withdraw, the water
molecules from the coats of cell components in the presence of Melafen are not
likely. On the contrary, to structure the bulk water and the neared membrane water
layers it is quite likely.
However, at present chapter, we are emphasized the attention to the Melafen aqua
solutions actions at the animal cellular level in vitro .
This is why, we tried to clear up the most possible speci¿ c targets for Melafen on
animal's cells surface. And several types of easily emitted cells of different etiology or
different origin were having picked up. The main task of work was the investigated the
Melafen action to the three types of cellular plasmalemmal receptors - purinoreceptors
P2Y, P2X, P2Z that present at the three types of cells--EAK cells, thymocytes, and
lymphocytes.
The Melafen is a melamine salt of bis (hydroximethyl) phosphinic acid. It is
a hydrophilic poly function substance with multi targets for its actions (Figure 1).
There are the phosphoric, hydroximethyl groups, and nitrogen contained structures at
Melafen molecule, potentially pointed to the certain targets at the biological cells. The
pretreatment of crop seeds by aqueous solutions of Melafen at concentration 10 -11 - 10 -9
M increased the yield of plant production by 11% or more due to the increasing of
plants stress tolerance under the bed environment [10]. But the increasing of the con-
centration of Melafen to 10 - 7 M and higher inhibits the processes of plants growth
completely [19]. Therefore, studies were carried out at a wide range of concentrations
(10 -21 -10 - 2 M). The main purpose was to determine how the aqueous solutions of
Melafen in a wide range of concentrations inÀ uence to the function of animal cells in
vitro only.
