In-Depth Information
4. Notes
1. Which fi xative is better?
Any aldehyde can be used for electron microscopy. In general,
glutaraldehyde and/or paraformaldehyde are more popular
than formaldehyde in electron microscopy. If the rest of the
specimen is used for immunostaining of light microscopy,
glutaraldehyde should not be used.
2. How to produce the specimen for scanning electron micros-
copy without injuring the luminal surface?
Like Fig. 2 , the end of the artery is stitched, only the adventitia
of the artery is picked up with microtweezers, and the artery is
cut with scissors under an operative microscope.
