9. RNase A cleaves single-stranded RNA, which leads to unspecifi c
cleavage of the pre-miRNA beacon that could also induce fl uo-
rescent increase. To avoid false-positive results, test your assay
setup by including 0.3-0.8 U/μl
l RNase inhibitor in the reac-
tion mixture. The kinetics of fl uorescence increase should be
independent of the presence of RNAse inhibitor.
