15. When programming the Ettan DALT twelve , ensure that the
pump is turned on, and that a suitable temperature (for instance,
10°C) is used. A temperature of less than 10°C should not
be used.
16. It is possible to scan the colloidal Coomassie images using a
normal fl at-bed scanner. However, the process of matching the
Coomassie image to the DIGE image is simplifi ed if the
Typhoon scanner is used throughout.
17. The light box used for gel-picking should be placed in an ergo-
nomically advantageous position, and the environment should
be as clean and dust-free as possible. Any contamination that
is introduced after electrophoretic separation is likely to inter-
fere with the nanoLC-MS/MS analysis of the proteins.
