12. Trim off a small portion of the IPG strips at both ends to
provide space for the addition of molecular weight markers
if required. The molecular weight markers can be pipetted
onto a small piece of fi lter paper and allowed to air-dry prior
to introducing it onto the second dimension gel.
13. Two gels can be scanned simultaneously in a Typhoon variable
mode imager at approximately 10 min/channel using 100-mm
scan resolution. The remaining gels have to be protected from
light and kept moist before image acquisition can be performed.
14. Optimization of PMT voltage can be done by scanning a
smaller area at high resolution or the fi nal resolution intended.
In this way, we can save time and the pixel output is more rep-
resentative of the fi nal value than pixel output obtained from
1,000-mm scan resolution.
