FIGURE 12.6
Cellular consequences of Gsa-tubuiln interaction. (A) Coimmunoprecipitation of Gsa
with tubulin in cells. PC12 cells were infected with adenovirus containing GFP alone,
HA-Gsa
Q227L -GTP, or HA-Gsa (wild-type) cDNA. Thirty-six hours after infection, wild-type
Gsa-infected cells were treated with cholera toxin (CTX) for 1.5 h. Cells were lysed, and Gsa
was immunoprecipitated using an HA antibody. Western blots with anti-tubulin antibody
reveal the extent of tubulin complexed with the two forms of Gsa. (B) Activated Gsa promotes
neurite outgrowth in PKA-deficient PC12 cells (PC12-123.7). Cells were infected with
adenovirus containing both GFP and Gsa or Gsa
Q227L . Images were acquired after cells were
infected with indicated viruses. Lower: Fluorescence of GFP indicated infected cells
expressing Gsa. Upper: Differential interference contrast. Bar: 20 mm.
