clearance of the basic isoforms, but only serves to decrease product recovery. Following
the intermediate wash step, the product is recovered from the column with an elution
buffer of increased ionic strength.
During pilot-scale testing of the purification process, unanticipated variation in
product recovery was observed. Step yields were up to 20% lower than that predicted by
small-scale development studies. The yield loss was due to an atypically high loss of the
