size of the aquasomes. The results of both TEM ( Figure 7.5 ) and PCS indicated
that the aquasomes were about 47 ± 19 nm with a polydispersity of 0.32. The
OVA loading was estimated at 60 ± 2 ug/mg. 111
7.4.2.3   Adsorption Efficiency and OVA Release from Aquasomes
The percent efficiency of OVA adsorption on aquasomes is determined as follows 146 :
(1) Weigh out 200 mg of OVA adsorbed aquasome formulations and suspended
in Triton-X 100 (0.01%, w/v).
(2) Incubate in a wrist action shaker for 1 h.
(3) Centrifuge at 14,000 rpm for 15 min.
(4) Collect the supernatant and perform micro BCA method for protein analy-
sis using aquasome formulation treated in the same manner as blank.
(5) For in vitro release studies, take 200 mg of OVA adsorbed aquasomes and
suspend in phosphate buffer (pH 7.4, 10 mL) with slow magnetic stirring.
Withdraw 1 mL samples every 15 min followed by centrifugation (1700 rpm
for 10 min at 4 °C).
