Selection of optimal laser lines, fluorophores, and band-pass filter combinations for multicolor
single-molecule detection fluorescence experiments. Fluorophores for SMD experiments
should show high absorption and quantum yield, low frequency of intersystem crossing, and
high photostability. An attractive combination of lasers and fluorescent dyes for triple-color
SMD-TIRF experiments is shown. In addition, epifluorescence excitation by a 385 nm LED
facilitates localization of cell stained with nuclear stains (DAPI or Hoechst 33342). The 405 nm
laser line enables efficient excitation of single quantum dot fluorescent nanocrystals matched
to one or more emission bands. The spectra of Alexa 555 and Alexa 594 dyes illustrate the
trade-off to match the 561 nm excitation or to match the emission band around 605 nm.
