The transcription factor T H -inducing POZ/Kr uppel-like factor THPOK 114
is
required after initial lineage specification for the differentiation of CD4
+
helper
T cells from double-positive CD4
thymocytes. Overexpression of THPOK
(ZBTB7b) converts MHC class-1-restricted CD8
+
,CD8
+
+
T cells into MHC class-2-
restricted CD4
+
T cells, whereas THPOK repression redirects CD4
+
T cells to the
CD8
lineage commitmemt.
Regulator GATA3 acts upstream from THPOK to initiate CD4
+
lineage [ 227 ]. Regulator THPOK represses the CD8
+
+
specification,
whereas THPOK serves to stabilize the CD4
fate [ 228 , 229 ]. Factor GATA3 is
necessary for THPOK expression in double-positive thymocytes.
The development of CD8
+
cytotoxic T-cell requires Runx1 and Runx3 tran-
scription factors. Both CD4 and THPOK are inhibited in CD8
+
cytotoxic T cells.
Conversely, THPOK represses the expression of Runx3 and other molecules of
