in an osteogenic medium for three weeks, which was composed of
minimum essential medium- α (MEM- α ) supplemented with 10%
fetalbovineserum(FBS),0.1 μ Mdexamethasone(Sigma),10mm β -
glycerolphosphate(Sigma),and50 μ ML-ascorbicacid2-phosphate
(Sigma). Platelet-rich plasma (PRP) was used as a carrier for both
CB-MSC groups. The BMP-2 injected group(15 rats) was that E-
coli derived human recombinant BMP-2s(rhBMP-2, Cowell Medi,
Busan, Korea) were injected at distraction site. The control group
was not injected with any material. The CB-MSC-injected group
and the osteogenically primed CB-MSC group received an injec-
tion of 0.05 mL CB-MSC and osteogenically primed CB-MSC, respec-
tively, at the distracted site. The BMP-2 injected group received 0.05
mL(0.1mg/ml) at the distraction site.
In the osteogenically primed CB-MSC + PRP-injected group and
the BMP-2 injected group, bone regeneration was evident in 100%
of the animals, but in the control and the CB-MSC + PRP-injected
group, bone regeneration was completed in 50% of the animals at
fourweeksandtheosteogenicallyprimedCB-MSC + PRPhaspositive
effective bone regeneration at this early time point. However, more
than 50% bone regeneration was also found in the CB-MSC + PRP-
injectedgroupfromsixweeks.Furthermore,similaramountofbone
regeneration was estimated in the CB-MSC + PRP-injected group
andtheosteogenicallyprimedCB-MSC + PRP-injectedgroupandthe
BMP-2 injected group at eight weeks. The control group showed
delayed bone regeneration in comparison to the CB-MSC + PRP-
injectedgroupandtheosteogenicallyprimedCB-MSC + PRP-injected
group at eight weeks (Fig. 43.8). In the histological examination at
eight weeks, new bone formation of the control group was visible
at the distraction site, but new bone formation of the osteogenically
primed CB-MSC-injected group and the BMP-2 injected group was
more robust and had larger enchondral ossification layers than the
control group and the CB-MSC-injected group (Fig. 43.9). The CB-
MSC-injected group and the osteogenically primed CB-MSC + PRP-
injected group also had more osteocytes than the control group;
the size of the callus was compared through an image analyzer
(Table43.1).
From this study, osteogenically primed CB-MSCs and BMP-2
accelerated the consolidation of the distraction area in respect of
