In this chapter, the preparation of extracellular matrix (ECM)-
based scaffolds from scratch is described. For applications in soft
tissue engineering and regenerative medicine, scaffolds based on
ECM proteins like collagen and elastin can be prepared with both
a known architecture and a defined composition. In combina-
tion with appropriate modulating molecules, like growth factors,
this may yield scaffolds containing the correct cues and signals
to trigger regeneration of specific tissues in vivo . The construc-
tion of innovative scaffolds with a defined molecular composi-
tion, morphology and shape will be described in this chapter. The
