with an aminoethyl side chain, the grafting of the bioreducible PAMAM with
dendrimer greatly improved the transfection efficiencies of 293T and HeLa
cells with foreign DNA at various N/P ratios. Alternatively, Wu et al.
synthesized a serum-resistant PAMAM-based polypeptide dendrimer through
peptide bond linkages for gene delivery (Scheme 4.7B). 67 The obtained
compound, ALA, had nontoxic low-generation G2 PAMAM as its central
core, polyglutamate as its star-shaped backbone branches, and G1 PAMAM
as its branch grafts and peripheral terminals. ALA exhibited negligible
cytotoxicity, significant high transfection efficiency, exciting serum tolerant
capacity, and sustained transfection characteristics compared with commercial
G5 and Lipofectamine, and proved itself a promising and challenging potential
gene delivery carrier in safe and efficient in vivo gene delivery.
In addition to reducing cytotoxicity, various efforts have focused on
incorporating cell targeting capability into PAMAM-based dendriplexes.
