cationic lipids could induce effective gene transfection in HEK293 cells in
association with DOPE. The gene transfection efficiencies of two cationic
lipids were dramatically increased in the presence of calcium ion (Ca 2+ ), and
their gene transfection abilities were maintained in the presence of 10% serum.
4.6.2 Polyallylamine
Polyallylamine, another synthetic cationic polymer, possesses a high density of
primary amino groups which are suitable for binding and packaging negatively
charged DNA. However, the utility of polyallylamine for gene delivery
application is limited by its cytotoxicity of a too strong polycationic character.
Different chemical modifications have been used to decrease the cytotoxicity
and enhance the transfection efficiency of polyallylamine. For example,
Pathak et al. demonstrated that polyallylamine modified with imidazole and
PEG-bis(phosphate) could reduce the positive surface charge to decrease the
