28. When growing overnight cultures in a 96-well microtiter plate,
we add a sterile 5 mm diameter glass bead (Thermo Scientifi c,
11-312C) to each well and put the plate in a shaking incubator
to keep the cells agitated overnight. Do not add the bead
before pinning with a robot as this may crash the robot.
29. This will give you 2 × 8 glucose standards of 33.3, 22.2, 14.8,
9.88, 6.58, 4.39, and 0 nm glucose, respectively (i.e., 2/3
dilution down the plate).
30. It is best to do one plate at a time for the ease of timing all the
steps in sequence.
31. Incubation times may vary as OD 540 values need to be obtained
in the 0.05-0.3 range. Do a test run with a few strains and
controls to get the best incubation time.
