Cultivation of hMSC on plotted CPC scaffolds. Prior to cell seeding, the scaffolds were steril-
ized by gamma radiation and preincubated in cell culture medium for 24 hours. Seeding was
performed semidynamically by incubation of the scaffolds overnight in a cell suspension with
gentle shaking. Osteogenic differentiation of hMSC was induced by addition of 10
-7
M dexa-
methasone, 3.5 mM β-glycerophosphate, and 0.05 mM ascorbic acid 2-phosphate to the cell
culture medium. Cell morphology and distribution was observed by SEM on (a, b) day 6 and
(c, d) day 12. Proliferation and osteogenic differentiation were detected by measurement of (e)
cytosolic LDH activity and (f) specific ALP activity.
