Chromatography
Separation of constituents from the mixture has remained the main focal
point in chromatography. Chromatography is the collection of a set of
laboratory techniques. There are two phases—the mobile phase , which
carries, and the stationary phase . The various constituents of the mixture
travel at different speeds, causing them to separate. Chromatography may
be preparative (separate the components of a mixture) or analytical
(measuring the relative proportions of analytes in a mixture).
Chromatography may be categorized by (a) chromatographic bed shape
(column chromatography, planar chromatography, paper chromatography,
thin-layer chromatography) and (b) physical state of mobile phase (gas
chromatography, liquid chromatography, etc.)
HPLC
One of the important chromatographic techniques in analytical chemistry
and biochemistry is the high-performance liquid chromatography, also
known as high-pressure liquid chromatography (HPLC), used to separate a
mixture of compounds for identifying, quantifying, and purifying the
individual components of the mixture. HPLC has many uses in medical,
purifying substances from a complex biological sample, manufacturing,
pharmaceutical, quality assurance, etc.
