Furthermore, as the present site-selective scission proceeds completely via a
hydrolytic pathway, the scission fragments can be recombined with foreign DNA
to provide desired recombinant DNA. The fundamental processes of DNA manipula-
tion (site-selective scission of DNA, followed by enzymatic ligation) are successfully
accomplished. The methods presented here are satisfactorily compatible with current
molecular biology and biotechnology, and thus should be highly promising for future
applications. Such possibilities, as well as attempts to further improve scission-
efficiency and -selectivity, are currently being pursued in our laboratory.
