2. Remove milk bottles from refrigerator and allow them to come
to room temperature.
3. Four hours after injections, milk bottles are placed on each
cage in the place of the normal water for a 20-min period.
4. Milk bottles are then removed and normal water bottles are
returned to all cages.
5. Repeat injections and milk habituation the next day.
6. Discard milk solution.
1. EPO (500 U/kg) and vehicle are injected as on previous
days.
2. Test occurs in homecages in their usual locations in the normal
housing room. Prepare fresh milk solution and partially fill
clean bottles.
3. Four hours after the injections, the test is performed. Water
bottles are removed from the cages and are replaced with milk
bottles. A timer is started and the latency it takes each mouse
to drink the milk is recorded. More than one mouse can be
tested at a time.
4. Label bottles and refrigerate for use in novel-cage test.
3.3.2 Home-Cage
Consumption Test (Day 4)
1. EPO (500 U/kg) and vehicle are injected as on previous days.
2. All mice to be tested are brought in their homecages into a
quiet holding room near the testing room (see Note 6).
3. Remove bottles of milk from refrigerator and allow them to
come to room temperature.
4. Prepare testing room. Have in the room a stack of clean mouse
cages and tops equal to the number of mice to be tested. The
room should have even bright lighting that falls evenly into the
test area (see Note 7). Place two of the mouse cages without
bedding side by side so that the observer has a clear view of the
entire cage area. A white divider (cardboard or Plexiglas) is
placed between the cages.
5. The novelty test is performed 4 h after the injections. Two
mice at a time are brought from the holding room into the
testing room and one is placed in each cage (see Notes 3
and 5). Milk bottles are placed on the cages and the timer is
started (see Note 8).
6. The time when each mouse begins to drink the milk is
recorded.
7. The test is terminated after 8 min for any mouse that does not
drink and a cutoff latency of 480 s is given to that mouse.
8. Mice are removed from test cages and returned to homecages
when both mice have completed the test.
