Fig. 28.3 Knockdown of TauT by RNAi reduces 293 cell colony formation in soft agar. Viable
colonies were stained with 0.2% p-iodonitrotetrazolium violet ( Sigma ) and appear blue on the
dish. After a period of 2 weeks, colonies were counted ( left ). Seventy-two colonies were counted
in the mock culture, whereas only 25 were counted in the shRNA-TauT culture. Therefore, results
demonstrate that TauT inhibition decreases colony formation and causes prolonged wound-healing
time ( bottom )
These results demonstrated that a reduction in the taurine transporter protein
level decreased the ability of 293 cells to form colonies in soft agar. Also, a wound-
healing test showed that control cells repaired wounds within 48 h, while the TauT-
deficient cells showed a much slower wound repairing process and a prolonged
wound-healing time compared to the controls.
