As another interesting approach, Mendez-Ardoy et al. developed a family of
discrete and well-characterized amphiphilic polycationic
-CD “click” clusters as
gene delivery vehicles (Scheme 11.13). The cationic structure for DNA complexation
was first installed on the primary hydroxyl group of
-CD by “click” HDC reaction.
Hydrophobic tails were then conjugated to the secondary hydroxyl group. Such CD-
centered amphiphilic compounds could self-assemble and complex with pDNA. By
tuning the density of positive charges, their distance to the CDs' core and the length
