Fig. 2 ( a ) NeuN/GFP double staining after injection of a PD relevant promoter into
rat striatum. The promoter was based on the promoter of the ring fi nger protein
25 (RNF25) gene. This gene is highly expressed in parkinsonian brain compared
to healthy brain. The promoter was cloned into a lentiviral vector expressing GFP
and injected into striatum of rats for in vivo validation. Scale bars 50
m.
( b ) Histogram comparing the level of neuronal specifi city of the RNF25 promoter
to that of the commonly used NSE promoter [ 27 ]. ( c ) Histogram comparing
the effi ciency of the RNF25 vector with other lentiviral vectors expressing from
commonly used ubiquitous promoters [ 27 ]

vious studies [ 38 ].

Notes
1. UCSC genome browser (http://www.genome.ucsc.edu) can
be used to visualize regions that are conserved between species
upstream of the transcription start site. These conserved
regions may be important in the regulation of the gene and by
including many of these regions in the promoter construct a
similar transgene expression and cell specifi city may be achieved.
This approach is benefi cial when a longer promoter is going to
