Fig. 5 In Vivo Expression of CREBCA Enhances LTP but not LTD. ( a ) Summary
graphs of LTP elicited in uninfected ( n = 7), GFP-expressing ( n = 5) and CREBCA-
expressing ( n = 7) neurons. Traces show sample EPSCs pre- (1) and post- (2,
45-60). ( b ) Summary graphs of LTD elicited in uninfected ( n = 6), GFP-expressing
( n = 5) and CREBCA-expressing ( n = 5) neurons. Traces show sample EPSCs pre-
(1) and post- (2, 25-40 min post) LTD induction (Scale bars: 20 ms/20 pA)

Notes
During cloning of the recombinant protein within the virus
backbone, it is essential to optimize the ligation procedure by
determining the molar ends of both products (vector and insert)
and calculating the amount of each product to obtain an adequate
ratio. Details of this optimization are usually provided with the
ligation kit. Deviating from the optimal ratio will signifi cantly
decrease your success for obtaining the clone desired.
During production of viruses, it is essential to optimize the
conditions of transfection and collection of the viral particles for
high titers. These high titers are necessary for good in vivo infection.
