In-Depth Information
5. Injection cannula (e.g., Plastic one cat. no. C315I)
6. Mineral oil
7. PB intramedia polyethylene (PE) tubing (Fisher USA cat. no.
1417011B)
8. Hair clipper
9. One scalpel, two hemostats
10. Surgical suture kit (needle and thread) or Michel suture clips
11. Xylazine hydrochloride, bupivacaine, ketamine hydrochloride,
and buprenorphine hydrochloride
L SGE injection syringe or similar
product is important to optimize the injection system. This syringe
has a dual gauge needle, which permits tight fi tting of the PE tub-
ing (item 5) to avoid air leaks and loss of pressure. Use of items 4,
5, and 7 together is recommended to obtain reliable pressure for
microinjection. Two compounds of item 11 (ketamine and
buprenorphine) are classifi ed as potential substance of abuse and
their purchase is regulated in several countries. An offi cial authori-
zation might be necessary prior to purchase.

2.3 Electrophysio-
logical Recordings
1. Antivibration table with faraday cage (e.g., TMC or Newport
workstations)
2. Microscope with the following specifi cs (e.g. Slice scope from
Scientifi ca, BX51WI from Olympus, or Axio Examiner from
Zeiss):
(a) Fluorescence for detection of infected neurons
(b) ×4 and ×60 objective
(c) Camera for visualization of cells for whole-cell patch
clamping
3. Recording chamber with heated fl ow circulation for slices
(e.g., slice recording chamber from Scientifi ca or Warner
Instruments)
4. Solution heater system (e.g., Temperature controller from
Scientifi ca or Warner Instruments)
5. Fine micromanipulator for whole-cell patch clamping (second
micromanipulator required if doing dual whole-cell patch-
clamp recordings, see Fig. 4 ) (e.g., MP series from Sutter
Instruments or PatchStar from Scientifi ca)
6. Microelectrode amplifi er and data acquisition/analysis system
for electrophysiology recordings (e.g., Multiclamp series,
Digidata and PClamp X from Axon)
7. Coarse micromanipulator for stimulation electrode (e.g., MX4
from Narishige or LBM-7 from Scientifi ca)
8. Stimulus isolator for generation of stimuli (e.g., Iso-Flex from
A.M.P.I.)
