FIGURE 14.3 Assembly of the T3SS injectisome. Cartoon representation of the major steps
involved in the assembly of the T3SS injectisome. (A) Elements of the export apparatus assemble in
the inner membrane. (B) This assembly serves as a scaffold for the arrangement of the basal body
inner- and outer-membrane proteins. (C) The ATPase and related components assemble inside the
cytoplasmic side, allowing secretion. (D) The ruler, rod, and needle proteins are secreted through
the basal body, extending the needle. (E) A substrate specificity switch prevents further extension
of the needle, which is capped by the tip/filament component. Translocon proteins are secreted. (F)
A second substrate specificity switch, upon contact with mammalian cells, induces the formation
of the translocation pore and the secretion of effector proteins. IM - inner membrane, OM - outer
membrane, HM - host membrane.
