FIGURE 11.1 Flow diagram summarizing the sequencing and annotation of the Ascaris
suum genome, consisting of (1) procurement of adult female Ascaris suum , (2) extraction of
genomic DNA from adult female uterine tissue, (3) deep-sequencing on the Illumina Hi-Seq
Platform, (4) scaffolded assembly using SOAPdenovo, (5) RNA-sequencing of larval and
adult male and female transcripts to support gene predictions, (6) functional annotation
based on homology with key model organisms including Caenorhabditis elegans and
Drosophila melanogaster , (7) further functional annotation based on generalist (i.e. KEGG)
and specialist [e.g. MEROPs (peptidases) and KS-Sarfari (kinases)] protein databases, and
(8) prediction of druggability based on assessment of lethal phenotypes in homologous
genes (i.e. essentiality) in model organisms and/or identification of metabolic chokepoints.
