occupied by the producers? Are lasso peptides chemical weapons playing a role in
bacterial competitions, or rather regulatory factors that could act as peptide auto-
inducers, or both? Are novel chaperones involved in the precursor prefolding that
can help in the formation of the lasso topology? What is the role played in the bio-
logical functions by the lasso handedness, which is right-handed for all known lasso
peptides while there is approximately no difference in energy between the right- and
left-handedness, as pointed by Link and co-workers (Ferguson et al. 2010 ; Maksi-
mov et al. 2012a )? This brief textbook reviews the current knowledge on lasso
peptides' origins, structures, biological activities, structure-activity relationships,
biosynthesis, genome mining discovery and bioengineering.
